Unlocking the potential of natural products in drug discovery

Drug Target Review

Between 2000 and 2020, approximately 30 percent of the newly introduced small molecule drugs were derived from natural products. In recent decades there has been a decline in interest in natural products for drug discovery, with the industry gravitating towards screening libraries of synthetic molecules with predefined chemistries.

HSYA prevents IL?1??induced human chondrocyte damage in osteoarthritis by inhibiting NF??B activation

Chemical Biology and Drug Design

ROS production and levels of pro-inflammatory cytokines in C28/I2 cells treated with HSYA were measured by immunoflurescence stating and q-PCR. The viability of C28/I2 and iCell-0092a chondrocytes was measured by CCK8 assay at different concentrations (2.5, 10, 40, and 160 μM).

Biochemical assays and deep cyclic inhibition in cancer treatment

Drug Target Review

We are pleased that to date our lead product candidate has aligned DCI profiles as expected. In April 2023, we presented preliminary safety data at the AACR Annual Meeting where our lead product candidate was observed to be well tolerated with no dose limiting toxicities observed in cancer patients through the first four dose levels.

Finding the right CDMO partner for cell line development

Drug Target Review

However, there is significant variability within the various CHO cell lines, and CDMOs often have differing levels of success in optimising their cells for protein production. Skilled CDMOs can also locate or create favourable genomic microenvironments where transcription factors are efficiently recruited, further boosting protein production.

Malaria unravelled: decoding the parasite’s gene expression control

Drug Target Review

The first identification was based on a bioinformatic analysis. The ApSIGMA gene product, an apicoplast RNA polymerase sigma factor ApSig, is imported into the apicoplast after the translation in the parasite cytosol and activates transcription from the apicoplast genome.
