Coumarin-3-carboxamide analogues were designed, synthesized, and evaluated for pancreatic lipase (PL) inhibition. Among the analogues, 5dh and 5de showed best PL inhibition, with IC 50 values of 9.20 and 11.4M, respectively, compared to Orlistat (IC 50 =0.97M). Analogue 5dh inhibited PL competitively with a K i of 4.504M.
